What is Compound Glucoamylase?

 

1. The compound saccharification enzyme is a compound saccharification enzyme composed of glucoamylase and pullulanone from which transsidase is removed. Among them, glucoamylase is refined by submerged fermentation and refining of Aspergillus niger, and pullulanase is refined by submerged fermentation and refining of Bacillus licheniformis. It is widely used in beer, monosodium glutamate, starch sugar, antibiotics and other industries that require maximum conversion of starch into glucose.

2. Compound Glucoamylase, also known as glucoamylase, can hydrolyze the α-1,4 glucosidic bond of starch from the non-reducing end to produce glucose, and it can also slowly hydrolyze the α-1,6 glycosidic bond to convert it into linear polysaccharides. Pullulanase is a thermostable debranching enzyme (starch a-1.6-glucosidase) used at low pH. It can hydrolyze the a-1,6-D-glycosidic bond in liquefied starch to produce a protein containing 1, The linear polysaccharide with 4-α-D-glucosidic bond can act on amylopectin quickly and effectively, and efficiently cooperate with glucoamylase to accelerate the reaction speed and increase the glucose content.

1. Temperature range: The effective temperature range is 40-65℃, the most suitable temperature is 55-60℃.

2. pH range: the effective pH range is 4.0-5.5, and the optimum pH range is 4.2-4.6.

3. The effect of metal ions on enzyme activity: Mg can activate the enzyme; Cu\M²Fe’ has an inhibitory effect; Zn promotes the activity of pullulanase, but inhibits the activity of glucoamylase.

Compound Glucoamylase application

 

1. When producing glucose, after the raw material is liquefied, adjust the pH to 4.2-4.5, cool to 58-60℃, add compound glucoamylase, generally add 0.4-0.9L/t dry starch, saccharification time is 30-60 hour.

2. In industries with moderate starch conversion such as beer, the recommended dosage is 0.15-0.35L/t dry starch.
